BAM Media M3: Acetate Agar

Sodium acetate 2 g
NaCl 5 g
MgSO4 (anhydrous) 0.2 g
Ammonium phosphate 1 g
K2HPO4 1 g
Bromthymol blue 0.08 g
Agar 20 g
Distilled water 1 liter

Add all ingredients except MgSO4 to 1 liter distilled water. Heat to boiling with stirring. Add MgSO4 and adjust pH. Dispense 8 ml portions into 16 × 150 mm tubes. Autoclave 15 min at 121°C. Incline tubes to obtain 5 cm slant. Final pH, 6.7.

NOTE: This medium contains a slightly smaller concentration of acetate than the formula recommended in Ewing (1986).
