Proteose peptone No. 3 20 g
Glycerol, C.P. 10 ml
K2HPO4 1.5 g
MgSO4 1.5 g
Agar 15 g
Distilled water 1 liter

Add all ingredients except MgSO4. Heat with agitation to dissolve agar. Adjust pH to 7.2 ± 0.2. Add MgSO4 slowly and mix. Dispense 4 ml portions to 13 × 100 tubes. Autoclave 15 min at 121°C.

Incline tubes to give half butt and half slant.
