BAM Media M205: Cronobacter Selective Broth (CSB)

Formulation and preparation follow the protocol in ISO 22964: 2017.
Media base

| Meat extract | 3.0 g / liter |
| Sodium chloride (NaCl) | 5.0 g / liter |
| Bromocresol purple | 0.04 g / liter |
| Sucrose (C12H22O11) | 10.0 g / liter |
| Water | 1 liter |
Weigh ingredients and dissolve in water, by heating if necessary. Adjust the pH, if necessary, so that after sterilization the pH is 7.4 ± 0.2 at room temperature. Dispense 10 ml of the base medium into tubes. Autoclave at 121°C for 15 min. Store the base medium at 4°C for up to 6 months.
Vancomycin solution
Dissolve 10 mg vancomycin hydrochloride in 10 ml distilled water. Mix and filter-sterilize the solution. Store the solution at 4°C for up to 15 days.
Complete medium
Add 0.1 ml of vancomycin solution to 10 ml of base medium (final concentration of vancomycin 10 mg/liter of CSB). Store the complete CSB at 4°C and use within 1 day.
