Tryptone 10 g
Neopeptone 10 g
Agar 2 g
Sodium thioglycollate 0.25 g

Dissolve all ingredients except agar and adjust pH to 7.4 ± 0.2. Add agar and heat with agitation to dissolve. Dispense 9 ml portions into 16 × 125 mm tubes. Autoclave 15 min at 121 °C. Before use, heat in boiling water or flowing steam for 10 min. Add 1 ml of sterile 10% carbohydrate solution to 9 ml base.
