BAM Media M103a: Modified Oxford Listeria Selective Agar

Columbia Blood Agar Base (brand dependent) 39.0-44.0 g
Agar 2.0 g
Esculin 1.0 g
Ferric ammonium citrate 0.5 g
Lithium chloride (Sigma L0505 quality or equivalent) 15.0 g
Buffered colistin methane sulfonate (1 % w/v) solution 1.0 ml
Distilled water 1.0 L

Hydrate the basal medium powder by stirring gradually into distilled water. Adjust pH to 7.2±0.1 if need be. Autoclave at 121° C for 10 min. Mix again. Cool rapidly to 46 ° C in a waterbath. Add 2 ml of 1 % w/v filter sterilized buffered moxalactam solution and mix well. Pour 12 ml per Petri dish.

Buffered 1% w/v colistin solution:

Colistin methane sulfonate (Sigma C1511 quality or equivalent) 1.0 g
0.1 M Potassium phosphate buffer, pH 6.0±0.1 100 ml

Dispense buffered solution as 5 mL aliquots. Store aliquots at -20° C.

Buffered 1% w/v moxalactam solution:

Sodium (or ammonium) moxalactam (Sigma M1900 quality or equivalent) 1.0 g
0.1 M Potassium phosphate buffer, pH 6.0±0.1 100 ml

Dissolve, filter-sterilize, and dispense as 2-ml aliquots. Store at -20° C.
