Agar | 4 g |
Distilled water | 1 liter |
General Instructions: Heat with agitation and boil 1-2 min to dissolve agar. For use with Salmonella, see instructions listed below. Dispense 8 ml portions into 16 × 150 screw-cap tubes. Autoclave 15 min at 121°C. Final pH, 7.4 ± 0.2.
*For use with halophilic Vibrio spp., add NaCl to a final concentration of 2-3%.
For Listeria: Keep individual tubes tightly screw capped and sealed with parafilm. Store in refrigerator for up to two weeks.
For Salmonella: Dispense 20 ml portions into 20 × 150 mm screw-cap tubes, replacing caps loosely. Autoclave 15 min at 121°C. Cool to 45°C after autoclaving. Tighten caps, and refrigerate at 5-8°C. To use, remelt in boiling water or flowing steam, and cool to 45°C. Aseptically dispense 20 ml portions into sterile 15 × 100 mm petri plates. Cover plates and let solidify. Use same day as prepared. Final pH, 7.4 ± 0.2.
CAUTION: For Salmonella: USE SAME DAY AS PREPARED
